The match against the Northern Premier League Premier Division will get underway at 7.45pm at South Kesteven Stadium.
It's pay on the day there, and admission will cost £8 adults, £5 concessions and £2 U18s, while children aged under 14 can attend for free if accompanied by a paying adult.
The competition returns after not taking place last season due to the COVID-19 pandemic, with the Iron's last involvement coming in 2019 when a youthful side was knocked out by Boston United in Round One.
Scunthorpe are 20-time winners of the competition, but last picked up the accolade a decade ago in 2011.
Incidentally, the last time United met the Gingerbreads was in the 2001-02 final of the Lincolnshire Senior Cup on August 1, 2002 (held over from the previous season).
On that day, the Iron won 3-0 thanks to a brace from Paul Wheatcroft and a Martin Carruthers goal.
